In tempered scales, as all modern instruments are tuned in, all notes but the root note is slightly out of tune. This allows us to play chords and modulate between different keys. When using an untempered scale, all you can do is basically to play one note, and harmonize against it with another note, but those notes are then perfectly in tune and if you happen to sing it, the harmony will resonate physically in your body. The difference from the tempered harmony is quite noticable.
